    Previously identified plasmacytoma modifier locus Pctr1 (plasmacytoma resistance 1) was narrowed to a 1.5 cM interval on mouse chromosome 4 using a congenic line carrying DNA spanning Cdkn2a (42.7 cM) to D4Mit15 (42.6 cM) from resistant donor strain DBA/2 on a BALB/c susceptible background. This line is called C.D2-(Cdkn2a-D4Mit15)/Lgm and is estimated to contain 1.5 cM of DBA/2 DNA (which encompasses the Pctr1 locus) on an otherwise BALB/c background. This line is significantly more resistant to plasmacytoma development compared to BALB/c.

    The authors propose Cdkn2a as a candidate gene for Pctr1. Inbred strain C57BL/6 is resistant to pristine-induced plasmacytomas but animals homozygous for the Cdkn2atm2.1Rdp knockout allele are susceptible and display accelerated pristine-induced plasmacytomagenesis. In addition, the Cdkn2a sequence contains polymorphisms P18H and I51V in the p16INK4a protein and H72R in the p19ARF protein between susceptible strain BALB/cAnPt and resistant strain DBA/2N, respectively. Overexpression of the BALB/c allele of Cdkn2a appears much less efficient in arresting cell growth of plasmacytoma cell lines in vitro compared to the DBA/2 allele of Cdkn2a.
