Linkage analysis was performed on 186 animals from a (NZW/LacJ x SM/J)F2 intercross and 186 animals from a (C57BLKS/J x SM/J)F2 intercross to identify QTL associated with white blood cell (WBC) count, platelet count, and mean platelet volume. Animals were phenotyped at 8 weeks of age. Parental strains NZW/LacJ and C57BLKS/J exhibit increased values for all 3 quantitative traits compared to parental strain SM/J. 88 polymorphic markers spaced 10 cM - 30 cM apart were used for the genome scan. Six white blood cell count QTL (Wbcq1-Wbcq6) were identified, three platelet count QTL (Pltq1-Pltq3) were identified, and two mean platelet volume QTL (Mpvq1-Mpvq2) were identified.

Wbcq1 (white blood cell QTL 1), Wbcq4, and Wbcq5 mapped to mouse Chromosome 1 at 38 cM (71 Mb), 58 cM (94 Mb), and 82 cM (156 Mb), respectively. These loci were analyzed using additional markers and a total of 279 F2 animals. Wbcq1 is linked to D1Mit282 (LOD=7) with C57BLKS/J-derived alleles conferring increased WBC count with dominant inheritance. Igfbp2 (36.1 cM) and Igfbp5 (36.1 cM) are two potential candidate genes for Wbcq1. Previously identified QTL Pbbcp1 (48 cM) maps near Wbcq1. Wbcq4 is linked to D1Mit306 (LOD=4.3) with NZW/LacJ-derived alleles conferring increased WBC count with additive inheritance. Previously identified QTL Pbbcp2 (63 cM) maps near Wbcp4. Wbcq5 is linked to D1Mit227 (LOD=4.2) with NZW/LacJ-derived alleles conferring increased WBC count with recessive inheritance. Creg1 is a potential candidate gene for Wbcq5.The 95% confidence intervals for Wbcq1, Wbcq4, and Wbcq5 span 36 cM - 52 cM , 50 cM - 72 cM , 76 cM - 90 cM, respectively.

Pltq1 (platelet QTL 1) mapped to 24 cM (35 Mb) on mouse Chromosome 2 near D2Mit296 (LOD=3.5). The 95% confidence interval of Pltq1spans 14 cM - 42 cM. SM/J-derived alleles confer increased platelet count with recessive inheritance.

Wbcq2 (white blood cell QTL 2) mapped to 42 cM (89 Mb) on mouse Chromosome 3 near D3Mit142 (LOD=3.2). The 95% confidence interval of Wbcq2 spans 24 cM - 47 cM. SM/J-derived alleles confer increased WBC count with recessive inheritance. This locus interacts with Wbcq3 on chromosome 15. Animals homozygous or heterozygous for SM/J-derived alleles at Wbcq2 and homozygous (or heterozygous) for C57BLKS/J-derived alleles at Wbcq3 exhibit further increases in white blood cell count.

Pltq2 (platelet QTL 2) mapped to 36 cM (52 Mb) on mouse Chromosome 7 near D7Mit30 (LOD=3.3). The 95% confidence interval of Pltq2 spans 24 cM - 52 cM. NWZ/LacJ-derived alleles confer increased platelet count with dominant inheritance.

Mpvq2 (mean platelet volume QTL 2) mapped to 44 cM (92 Mb) on mouse Chromosome 8 near D8Mit211 (LOD=2.7). The 95% confidence interval of Mpvq2 spans 24 cM - 49 cM. SM/J-derived alleles confer increased mean platelet volume with recessive inheritance. Tuba2, Tuba6, and Nfe2 (61.7 cM) are potential candidate genes for Mpvq2.

Pltq3 (platelet QTL 3) mapped to 10 cM (23 Mb) on mouse Chromosome 12 near D12Mit182 (LOD=3.2). The 95% confidence interval of Pltq3 spans 0 cM - 31 cM. NZW/LacJ-derived alleles confer increased platelet count with additive inheritance.

Wbcq3 (white blood cell QTL 3) mapped to 0 cM (0 Mb) on mouse Chromosome 15 near D15Mit13 (LOD=2.8). The 95% confidence interval spans 0 cM - 52 cM. SM/J-derived alleles at Wbcq3 confer increased WBC count with dominant inheritance. Mpvq1 (mean platelet volume QTL 1) mapped to 62 cM near D15Mit16 (LOD=14.2 in the NZSM cross; LOD=24.2 in the KSSM cross). The 95% confidence interval of Mpvq1 spans 58 cM - 60 cM. SMJ-derived alleles confer increased mean platelet volume with additive inheritance in the NZSM cross and recessive inheritance in the KSSM cross.

Wbcq6 (white blood cell QTL 6) mapped to 8 cM (24 Mb) on mouse Chromosome 14 near D14Mit98 (LOD=2.8). The 95% confidence interval spans 0 cM - 20 cM. Animals homozygous for NZW/LacJ- or SM/J-derived alleles at Wbcq6 exhibit increased WBC counts compared to heterozygous animals. Il3ra (1.5 cM) is a potential candidate gene for Wbcq6.
