Linkage analysis was performed to identify anxiety and neuroticsm loci in 751 animals from a heterogeneous stock derived from 8 different inbred strains (A/J, AKR, BALB/c, C3H, C57BL/6, DBA, I, and RIII. A QTL interval on mouse Chromosome 1 mapped to 64.84 cM near D1Mit394. This locus is near the previously identified emotionality QTL Emo3 at 73 cM, and the region is syntentic to human Chromosome 1 at 201 cM - 203 cM. Human neuroticism maps to Chromosome 1 at 113 cM - 144 cM.

The QTL peak spans a 0.8 cM(4.78 Mb) interval containing 17 pseudogenes and 9 genes. Sequence analysis revealed conservative coding variants in Glrx2 (77.8 cM), Trove2 (formerly Ssa2; 78.7 cM), and Rgs1 (78 cM). Two other potential candidate genes located in the QTL interval are Rgs2 (78 cM), and Rgs13 (78 cM). Complementation test was performed by crossing Rgs2 knockout animals with high-anxiety or low-anxiety inbred strains. Interaction between Rgs2 strain variant alleles and the knockout allele was observed.
