Linkage analysis was performed on 200 female animals from a (NFR/N x B10.Q-H2q)F1 x B10.Q-H2q backcross to identify loci associated with collagen-induced arthritis traits. 125 polymorphic markers were used for the genome scan. Female animals used in the study were aged (10 months) and induced with collagen after at least 4 possible pregnancies. Parental strain NFR/N exhibits delayed onset of and lower incidence of disease after collagen induction compared to parental strain B10.Q-H2q.

Significant linkage to CIA incidence (LOD=4.1) and onset (LOD=4.0) mapped to 64 cM - 70 cM on mouse Chromosome 11. This locus also shows suggestive linkage to arthritis score (LOD=3.3) and is named Cia40 (collagen induced arthritis 40). Markers flanking Cia40 are D11Mit360 (64 cM) and D11Mit214 (70 cM). The QTL peak occurs near D11Mit333 (66 cM) and D11Mit181 (69 cM). Previously identified QTLs mapping near Cia40 include Eae22 (61 cM) and Eae7 (48 cM). A QTL associated with pregnancy failure mapped using the same cross, Pregq2, coincides with Cia40.

Suggestive linkage to CIA incidence mapped to 27 cM on mouse Chromosome 1 between D1Mit373 (17 cM) and D1Mit216 (49.7 cM). Ctla4 (30 cM) is a potential candidate gene for this locus.

Suggestive linkage to anti-CII antibody titer mapped to mouse Chromosome 13 between D13Mit24 (43 cM) and D13Mit51 (59 cM). Previously identified QTL Cia28 (53 cM) maps near this locus. Authors speculate Cia28 is identical to the suggestive QTL at D13Mit24. However Cia28 was mapped using different parental strains (DBA/1J and FVB/N).
