    High and low alcohol preference selection lines HAP1 and LAP1 (and replicate lines HAP2 and LAP2) are derived from inbred strain HS/Ibg. At the 12th generation of selective breeding, parental strain HAP1 consumes on average 12 g/kg/day of 10% alcohol solution whereas parental strain LAP1 consumes on average 1.5 g/kg/day.

    Phenotypically extreme HAP1/LAP1 animals (n=96) and HAP2/LAP2 animals (n=48) were screened for microsatellite markers in chromosomal regions previously reported to influence alcohol preference phenotypes. Linkage to alcohol preference, Alpq1, mapped to chromosome 9 near D9Mit4 (29 cM) in the HAP1/LAP1 set.

    1.19.2016 Curator Note: Because Alpq1 was originally mapped here using the HAP1/LAP1 mice which differ from the HAP2/LAP2 mice, we consider the analysis of the HAP2/LAP2 mice a separate mapping experiment and have named the QTL identified Aplq9 .

    Alpq9 mapped near D9Mit90 (9 cM) in the HAP2/LAP2 set.

    The Alpq1 QTL interval is broad and may contain more than 1 underlying gene. Drd2 at 28 cM is a potential candidate for Alpq1. A previously identified alcohol QTLs Etohc3 (29 cM) and Ap5q (26 cM) map near this locus.

    Linkage to alcohol preference was also detected at D2Mit37 (45 cM) on mouse Chromosome 2 in the HAP2/LAP2 set. This locus is named Alpq2 (alcohol preference QTL 2). Scn1a (36 cM) and Nr4a2 (34.5 cM) are potential candidate genes near Alpq2.

    In addition, linkage analysis was performed on 432 (HAP1 x LAP1)F2 intercross animals using 134 microsatellite markers at an average resolution of 11.2 cM. A female-specific locus on mouse Chromosome 9 between D9Mit2 (17 cM) and D9Mit355 (53 cM) shows significant linkage to alcohol preference with LOD=5.04 and accounts for 11% of the variance. This QTL is named Alpq3 (alcohol preference QTL 3). A potential candidate gene for Alpq3 is Htr1b at 46 cM.

    Suggestive linkage to alcohol preference mapped to 107 cM on mouse Chromosome 2 (LOD=2.32 between D2Mit113 and D2Mit266) and 44 cM on mouse Chromosome 5 near D5Mit201 (LOD=2.93). Alcp5 and Alcp6 are previously identified alcohol QTLs mapping near the chromosome 2 locus. Gabra2, Gabrb1, and Gab2g1 at 40 cM are GABA receptor genes mapping near the chromosome 5 locus.
