    In silico SNP linkage disequilibrium analysis was performed to identify genomic loci associated with myeloid progenitor cell proliferation frequencies in response to cytokine stimulation, a precursor to acute myelogenous leukemia. Femur bone marrow from male mice at 8-12 weeks of age was used for colony-forming cell assays. Five inbred strains were examined. DBA/2J exhibits the highest level of myeloid progenitor cell proliferation in response to IL-3 and GM-CSF compared to C57BL/6J, AKR/J, C3H/HeJ, and BALB/cJ. The 5 strains did not differ much in response to SCF stimulation.

    Linkage to cytokine-stimulated myeloid progenitor cell proliferation mapped to a large interval on mouse Chromosome 3 from about 20 cM - 80 cM. The estimated smallest interval is30 cM - 40 cM and shows strong correlation to GM-CFS response. This locus is named Mpcp1 (myeloid progenitor cell proliferation 1). Potential candidate genes in this QTL interval include Csf1 (51 cM) and Pdgfc.

    Significant linkage to SCF-stimulated myeloid progenitor cell proliferation mapped to 20 cM - 50 cM on mouse Chromosome 4. This locus is named Mpcp2 (myeloid progenitor cell proliferation 2). Tal1 (49.5 cM) is a potential candidate gene mapping near Mpcp2. The TAL1 human ortholog is associated with leukemia in humans.

    Significant linkage to GM-CSF-stimulated myeloid progenitor cell proliferation mapped 10 cM - 40 cM on mouse Chromosome 5. This locus is named Mpcp3 (myeloid progenitor cell proliferation 3). A second interval, named Mpcp4 (myeloid progenitor cell proliferation 4), mapped to 40 cM - 78 cM. Lprm4 (54 cM) is a previously identified QTL mapping near Mpcp4. A potential candidate gene is AI586015.

    Mpcp5 (myeloid progenitor cell proliferation 5) mapped to 50 cM - 72 cM on mouse Chromosome 7 in linkage to GM-CSF response. Rflat1 and Fgf4 (72.4 cM) are potential candidate genes mapping near Mpcp5.

    SCF response mapped to 10 cM - 40 cM on mouse Chromosome 8. This locus is named Mpcp6 (myeloid progenitor cell proliferation 6). Dnase2a (38.6 cM) and Irf8 (65 cM) are potential candidate genes near Mpcp6.

    Significant linkage to IL-3 response mapped to 40 cM - 70 cM on mouse Chromosome 11. This locus is named Mpcp7 (myeloid progenitor cell proliferation 7). Apoh at 63 cM is a potential candidate gene mapping near Mpcp7.

    Significant linkage to SCF response mapped to a large interval spanning 10 cM - 50 cM on mouse Chromosome 13. This locus is named Mpcp8 (myeloid progenitor cell proliferation 8). Sfxn1 at 35 cM is a potential candidate gene.

    Mpcp9 (myeloid progenitor cell proliferation 9) mapped to 0 cM - 30 cM on mouse Chromosome 14 in linkage to IL-3 response. Il3ra at 1.5 cM is a potential candidate gene for Mpcp9. A locus at 40 cM - 70 cM, named Mpcp10 (myeloid progenitor cell proliferation 10), shows significant linkage to GM-CSF response. Tnfsf11 at 45 cM is a potential candidate gene for Mpcp10.

    Significant linkage to IL-3 response mapped to 20 cM - 60 cM on mouse Chromosome 17. This locus is named Mpcp11 (myeloid progenitor cell proliferation 11). Notch4 (18.7 cM), Brd4 (20 cM), and Runx2 (28 cM) are potential candidate genes.

    Significant linkage to GM-CSF response mapped to 30 cM - 58 cM on mouse Chromosome X. This locus is named Mpcp12 (myeloid progenitor cell proliferation 12). Mllt7 (36 cM), Il2rg (38 cM), and Btk (51 cM) are potential candidate genes. The human ortholog MLLT7 is associated with leukemia in humans.
