QTLs modifying Salmonella susceptibility locus Slc11a1 were identified using phenotype selection lines AIRmax and AIRmin. These lines were derived from 8 inbred strains and selected for maximal acute inflammatory reaction (AIRmax) or minimal acute inflammatory reaction (AIRmin). Salmonella resistance (RR) and susceptibility (SS) alleles at Slc11a1 became fixed in these selection lines and are referred to as AIRmaxRR, AIRmaxSS, AIRminRR, and AIRminSS. Microsatellite analysis identified 4 chromosomal region exhibiting allele fixation associated with susceptibility to S. tyhpimurium.

AIRmaxRR and AIRminRR animals displayed increased acute inflammatory reaction compared to AIRmaxSS and AIRminSS animals. Also, sensitivity to Salmonella infection is significantly increased in the AIRmaxSS line compared to the AIRminSS line. This QTL on mouse Chromosome 1 is named Msts1 (modifier of Salmonella typhimurium susceptibility 1). Slc11a1 (39.2 cM) is an obvious candidate gene for this locus. Il8rb (40 cM) is another potential candidate gene for Msts1. A previously identified Salmonella susceptibility QTL named Ssta1 (34.8 cM) maps near Msts1.

Msts2 mapped to 32.5 cM on mouse Chromosome 6 near D6Mit188 (P<0.0001). Potential candidate genes forMsts2 include Cd8a (32.5 cM), Il12rb2 (30.1 cM), and Igk (30 cM). Msts2 colocalizes with a previously identified Salmonella susceptibility QTL named Ssta3 (35 cM).

Msts3 mapped to 37 cM on mouse Chromosome 11 near D11Mit5 (P<0.0001). This locus colocalilzes with a previously identified Salmonella susceptibility QTL named Ssta4 (37 cM). Potential candidate genes mapping near this locus include Csf3 (57 cM), Csf2 (29.5 cM), Nos2 (45.6 cM), Mpo (49 cM), Il3 (28.5 cM), Il4 (29 cM), Il5 (29.2 cM), and Il13 (29 cM). Msts3 is syntenic to a region on human Chromosome 5q31-q33 associated with resistance to several diseases.

Msts4 mapped to 19 cM on mouse Chromosome 13 near D13Mit117 (P<0.0001). Tcrg (10 cM) is a potential candidate gene for Msts4.
