    A congenic line carrying the tuberculosis susceptibility QTL Sst1 was created by introgressing a 20 cM region of C57BL/6J-derived DNA from 43 cM - 64 cM on chromosome 1 onto a C3HeB/FeJ genetic background for 10 generations. This line is called C3H.B6-Sst1. The congenic was then crossed to C3SnSmn.CB17-Prkdcscid/J to create C3H.B6-Sst1 Pkrdcscid animals. Experimental animals were inoculated with 1 x 104 CFU of Listeria monocytogenes by tail vein injection. Infected congenic C3H.B6-Sst1 animals display 3-8 times decreased bacterial load in the liver and spleen compared to C3HeB/FeJ controls, indicating the C57BL/6J-derived Sst1 allele confers listeriosis resistance on a C3HeB/FeJ background. The C3H.B6-Sst1 Pkrdcscid animals show an even more dramatic effect with 500-1000 times decreased liver and spleen bacterial load compared to C3SnSmn.CB17-Prkdcscid/J controls. Infected C3H.B6-Sst1 Pkrdcscid animals also have significantly decreased necrotic lesions in the liver and spleen compared to C3SnSmn.CB17-Prkdcscid/J. B and T lymphocyte deficiency in C3H.B6-Sst1 Pkrdcscid animals was not affected by the congenic Sst1 interval.
