Linkage analysis was performed using 21 SMXA (SM=SM/J; A=A/J) recombinant inbred (RI) strains to identify QTLs associated with molar crown diameter. A total of 226 mice were phenotyped at age 40 days and a panel of >1000 polymorphic markers were used forgenome scan.

Significant linkage to buccolingual crown diameter for upper first molars in males mapped to 45 cM on mouse Chromosome 13 near D13Mit191 (LOD=3.54). This locus, designated Mcdq1 (molar crown diameter QTL 1), also exhibits suggestive linkage to mesodistal crown diameter for upper and lower first molars in both sexes. SM/J-derived alleles at Mcdq1 confer increased molar diameter with additive inheritance while A/J-derived alleles confer decreased diameter. Hexb at 46 cM is a potential candidate gene for Mcdq1.

Several suggestive linkages were detected on mouse Chromosomes 3, 7, 8, and 15. Linkage to buccolingual crown length for lower first molars mapped to 11.2 cM on mouse Chromosome 3 near D3Mit203 (LOD-2.61-3.24). Linkage to mesodistal crown diameter for upper first molars mapped to 51.4 cM on mouse Chromosome 7 near D7Mit17 (LOD=2.28-2.37). On the same chromosome linkage to mesodistal crown diameter for lower first molars mapped to 16 cM near D7Mit79 (LOD=2.13-2.28). On mouse Chromosome8, linkage to buccolingual crown diameter for upper first molars mapped to 67 cM near D8Mit14 (LOD=2.39-2.8). On mouse Chromosome 15, linkage to buccolungual crown diameter for lower first molars mapped to 32 cM near D15Mit17 (LOD=2.37-2.69).
