Sle16 is a previously identified susceptibility locus for systemic lupus erythematosus (SLE) mapping to distal mouse Chromosome 1. Congenic animals carrying a 129S6/SvEvTac-derived (susceptible) Sle16 locus on a C57BL/6 (resistant) genetic background develop humoral autoimmunity. Subcongenic lines were constructed to refine Sle16. Female animals were assessed for lupus traits at 6- and 12-months of age. The Sle16 critical region was narrowed to an interval 7.7 cM interval between D1Mit36 (92.3 cM, 92.3 Mb) and D1Mit358 (100 cM, 176.5 Mb). This interval is associated with autoantibody production with 129S6/SvEvTac-drived alleles conferring increased antibody production. The SLAM gene cluster mapping near Sle16 were suggested as possible candidates.
