Tlln is a tail length QTL previously identified in crosses between C57BL/6J and DBA/2J, and maps to 50 cM on mouse Chromosome 1. Tlln appears to have a pleiotropic effect on several skeletal phenotypes. However, it is unclear if the pleiotropic effect results from several closely-linked QTLs or is one major QTL.

The current study isolates a C57BL/6J-derived 0.94 Mb four-gene region of chromosome 1 on a DBA/2J genetic background. The congenic interval spans markers D1Icp1 (158.18 Mb) and D1Icp9 (159.12 Mb). Animals were sacrificed at 6- or 10-weeks of age after measuring tail length and 11 other skeletal phenotypes.

The refined Tlln congenic region segregates with tail length and three measures of mandible size, but does not show linkage to other skeletal phenotypes. Thus, it appears the original Tlln interval is composed of multiple linked QTLs. Consistent with previous studies, C57Bl/6J-derived alleles at Tlln confer increased tail length with additive inheritance. Pappa2 is one of the four genes present in the Tlln target interval and is considered a strong candidate gene.
