Small effect QTLs for collagen-induced arthritis (CIA) were mapped in 290 (DBA/1J x FVB/NJ)F2 mice. Since Cia2 was detected as a major locus in a previous study the data was analyzed while controlling for the Cia2 candidate gene C5. Animals were immunized with bovine collagen II at 8-12 weeks of age. Parental strain DBA/1J is susceptible to CIA compared to parental strain FVB/NJ. A panel of 126 polymorphic markers at a resolution of 11.5 cM was used for genome scan.

Linkage to CIA susceptibility mappedto 8 cM (23.9 Mb) on mouse Chromosome 5 near D5Mit349 (P=0.00691). This locus is named Secia1 (small effect CIA locus 1). Steap4, Add1 (20 cM), Wdr1 (24 cM), and Cd38 (28 cM) are potential candidate genes exhibiting differential expression between DBA/1Jand FVB/NJ. The QTL interval of Cia13 (34 cM) identified by a different group overlaps with Secia1. A second small effect locus named Secia2 was identified at 65 cM (119.1 Mb) near D5Mit425 (P=0.04397). Potential candidate genes for Secia2 include Tgfbr3, Oas1a (67 cM), Baz1b, Lfng (80 cM), and Gna12 (82 cM). This locus overlaps with previously identified Cia14 and is syntenic to regions on human Chromosomes 22q11 and 12p13-q24. DBA/1J-derived alleles at Secia1 and Secia2 confer CIA susceptibility.

FVB/NJ-derived CIA susceptibility mapped to 71.2 cM (145.7 Mb) on mouse Chromosome 6 near D6Mit14 (P=0.00315). This locus is named Secia3 (small effect CIA locus 3) and is syntenic to a region on human chromosome 12p13-pter. Etnk1 (74 cM) and 8430419L09Rik arepotential candidate genes for Secia3. These genes exhibit differential expression in DBA/1J and FVB/NJ.

A small effect CIA locus, Secia4, was detected at 27.8 cM (59.3 Mb) on mouse Chromosome 7 near D7Mit248 (P=0.0325). DBA/1J-derived alleles at Secia4 confer CIA susceptibility.

Secia5 mapped to 47 cM (85.1 Mb) on mouse Chromosome 10 near D10Mit261 (P=0.00647). DBA/1J-derived alleles at Secia5 confer CIA susceptibility. Rrp1 at 42 cM (formerly Nnp1) is differentially expressed in DBA/1J and FVB/NJ and isconsidered a potential candidate gene. The QTL interval of Cia8 (40.4 cM), a previously identified QTL, overlaps with Secia5. This locus is also syntenic to regions on human Chromosome 21q22-qter and 10q22-23.

Linkage to DBA/1J-derived CIA susceptibility mapped to 70 cM (115.2 Mb) on mouse Chromosome 11 near D11Mit214 (P=0.00521). This locus is named Secia6 (small effect CIA locus 6). Potential candidate genes for Secia6 differentially expressed between DBA/1J and FVB/NJ include Kpnb1, Stat5a (60.5 cM),and Pscd1. Secia6 overlaps with the Pgia7 (42 cM) QTL interval and is syntenic to a region on human Chromosome 17q21-q25.

Secia7 (small effect CIA locus 7) mapped to 31 cM (45.5 Mb) on mouse Chromosome 16 near D16Mit138 (P=0.01499). FVB/NJ-derived alleles at this locus confer CIA susceptibility. Secia7 is syntenic to a region on human Chromosome 3q29-qter.

    Secia8 (small effect CIA 8) mapped to 9.65 cM (18.3 Mb) on mouse Chromosome 17 near D17Mit197 (P=0.02994). FVB/NJ-derived alleles at Secia8 confer CIA susceptibility. Potential candidate genes for Secia8 include Mapk8ip3 (10 cM), Mapk14 (13.5 cM), A430107D22Rik, Hspa1a (18.9 cM), H2-Q10 (19.22 cM), and Gabbr1 (20.4 cM). These genes are differentially expressed between DBA/1J and FVB/NJ.
