    Linkage to snout length was previously mapped to mouse Chromosome 12 in a (C57BL/6J x DBA/2J)F2 intercross population (n=110). Parental strain C57BL/6J displays a longer snout and narrower face compared to parental strain DBA/2J. Linkage peaked at 44 cM near D12Mit7 and is named Ssnt1 (short snout 1). In this study, congenic line analysis was used to confirm Ssnt1.

    B6.D2-(D12Mit269-D12Mit134) and B6.D2-(rs3689063-rs3679276) are congenic lines designated B6.D2.12C and B6.D2.12M by the authors, respectively. Both lines carry DBA/2J-derived donor DNA containing the Ssnt1 locus on a C57BL/6J genetic background. Animals from both congenics displayed significantly shorter snout length and greater face width compared to C57BL/6J controls. A separate congenic B6.D2-(D12Mit269-D12Mit76) designated B6.D2.12P by the authors does not carry a DBA/2J-derived Ssnt1 locus and displays a longer snout and narrower face compared to 12C and 12M.

    This study appears to confirm a locus around 44 cM on mouse Chromosome 12 that impacts craniofacial development. Other modifying genetic factors may exist on chromosome 12 since congenic lines 12C (6 cM - 59 cM) and 12M (22 cM - 59 cM) both carry Ssnt1 but 12C animals tended towards shorter snouts, even shorter than that of parentalstrain DBA/2J.
