    Modifiers of susceptibility to collagen-induced arthritis (CIA) were mapped using 113 animals from a (DBA/1J x SWR/J-Tg(TcrbBL17)1Lmor)F2 intercross. Parental strain SWR/J-Tg(TcrbBL17)1Lmor carries the Tcrb transgene and is resistant to CIA compared to parental strain DBA/1J. F2 animals homozygous or heterozygous for the transgene had a 58% incidence of CIA with widely variable severity and onset.

    The Hc locus on chromosome 2 has previously been implicated in CIA susceptibility. Parental strain SWR/J carries Hc-null alleles (Hc0) while DBA/1J carries Hc-sufficient alleles. The author investigated Hc genotype in (DBA/1J x SWR/J-Tg(TcrbBL17)1Lmor)F2 transgenic animals and found that all CIA-affected transgenic animals had at least 1 copy of the functional DBA/1J Hc allele. Hc-deficient F2 animals, either transgenic or non-transgenic, were protected against CIA. However, not all Hc-sufficient transgenic F2 animals developed CIA suggesting the involvement of a second modifier QTL.

    Confirmation of Hc involvement in CIA was performed by genotyping 8 polymorphic markers on chromosome 2 in transgenic and non-transgenic F2 animals. The highest linkage for CIA susceptibility mapped to the Hc locus between D2Mit239 and D2Mit61 with a LOD score of 7. The authors indicate they do not exclude the possible involvement of other genes near Hc in CIA susceptibility.
