    A suppressor of SryPos sex-reversal mapped to mouse Chromosome 11 using a phenotype-driven congenic selection approach. Male hybrids of C57BL/6J and Mus domesticus poschiavinus experience sex reversal when carrying YPos sex chromosomes and display ovarian tissue development instead of testicular tissue development. Sex reversal is not observed when DBA/2J or 129S1/SvImJ is crossed to Mus domesticus poschiavinus indicating YPos compatible factors or suppressors of sex reversal are present in these two inbred strains.

    Consomic strain C57BL/6J-YPos was repeatedly back-crossed to 129S1/SmIvJ (a sex reversal-resistant strain) while selecting for phenotypically normal males in each generation until N13 resulting in B6.129-YPos congenic animals. Genome scan was then performed to identify 129S1/SvImJ-derived chromosomal segments conferring protection against sex reversal.

    A region of chromosome 11 between rs3685856 (14.267 Mb; 3 cM) and rs3088940 (56.329; 32 cM) is derived from 129S1/SvImJ. In addition, the region between rs13481176 (97.413; 57 cM) appears fragmented due to several recombination events. Some of the genetic material in this interval is neither C57BL/6J nor 129 in origin. However, authors consider this segment part of the B6.129-YPos congenic interval. This locus is Rsxr (resistance to sex reversal), originally published by Whitney et al, 2000. 129S1/SvImJ-derived alleles at Rsxr confer protection against sex reversal in B6.129-YPos animals. Homozygosity for 129S1/SvImJ-derived alleles at Rsxr confers a greater incidence of testicular tissue compared to Rsxr heterozygous or homozygous B6.129-SryPos animals.

    Several potential candidate genes are located within the chromosome 11 congenic region. Potential candidate genes displaying mRNA expression in the reproductive organs or urogenital ridge include Srebf1, Ncor1, Trp53, Shbg (35 cM), Sox15 (39 cM), Dvl2 (36.5 cM), Foxn1 (45 cM), Nle1, Lhx1 (48 cM), Tbx2 (49 cM) and Tbx4 (49 cM).
