Linkage analysis was performed on 1040 (LG/J x SM/J)F2 animals to identify genetic loci associated with skeletal traits. The F2 population consists of two sets of intercrosses where intercross I (n=533) was phenotyped at 10-weeks of age and intercross II(n=507) was phenotyped between 72- and 237-days of age. Parental strain LG/J displays increased body weight compared to SM/J. Seventy skeletal traits were measured. Intercross I was genotyped for 76 microsatellite markers on 19 autosomes (excluding X chromosome). Intercross II was genotyped at 95 microsatellite markers and 369 SNP markers, including 16 SNPs on chromosome X. One hundred five pleiotropic loci were identified representing 798 single trait QTLs. The average number of traits associated per QTLwas 7.6. In general, LG/J-derived alleles confer increased skeletal trait measurements.

    On mouse Chromosome 17, four skeletal traits QTLs were identified at 4.8 cM (Plskt82; LPR=4.02), 22 cM (Plskt83; LPR=21.56), 30 cM (Plskt84; LPR=5.75) and 71 cM (Plskt85; LPR=3.65).

A locus at 4.8 cM named Plskt82 (pleiotropic skeletal traits 82) displays linkage to six skeletal traits including face length in females, sacral length and width, eighth thoracic length, forelimb infraspinous length and fourth lumbar length. The Plskt82 confidence interval spans 0 cM to 17 cM.

Plskt83 (pleiotropic skeletal traits 83) is located at 22 cM and the QTL confidence interval spans 16 cM to 27 cM. This locus is associated with seventeen skeletal traits including hindlimb innominate length, first and fourth lumbar lengths, humerus length, mandible square root of coronoid process area in males, ulna length, femur length, tibia length, ilium length, palate length, face width, first lumbar centrum height in females, interorbital width, sacroilium joint length, eighth thoracic length in males and mandible square root of molar alveolus area.

Plskt84 (pleiotropic skeletal traits 84) mapped to 30 cM and exhibits linkage to six skeletal traits- third caudal centrum height, eighth thoracic length, mandible square root of masseteric area and square root of condyloid process area, skull frontal bone length and body weight (in females) at time of necropsy. The confidence interval for Plskt84 spans 24 cM to 36 cM.

A locus at 71 cM designated Plskt85 (pleiotropic skeletal traits 85) displays linkage to eight skeletal traits. The QTL confidence interval spans 29 cM to 76 cM. Measurements for third metatarsal length and third and eighth thoracic lengths are linked to Plskt85 in females only.
