Experiment
A QTL-rich region was identified on mouse Chromosome 1 spanning 172 Mb (92.3 cM) to 178 Mb (100 cM). This locus is named Qrr1 and contains approximately 32 previously identified QTLs for various traits including alcohol dependency, escape latency and emotionality. The borders of Qrr1 are defined by Fcgr3 (92.3 cM) and Rgs7 (99.3 cM). Expression phenotypes were examined for the Qrr1 region using 18 different mRNA datasets from 6 different mouse crosses. Fifty-two trans-QTLs mapped to the Qrr1 region in the BXD hippocampus and B6C3HF2 brain datasets. Twenty-six cis-QTLs with LOD>3 mapped to the Qrr1 region using the BXD CNS datasets. DBA/2J-derived alleles at Qrr1 confer increased expression of Sdhc, Ndufs2, Adamts2, Dedd, Pfdn2, Ltap, Pea15, Atp1a2, Kcnj9, Kcnj10, Igsf4b and Grem2. C57BL/6J-derived alleles at Qrr1 confer increased expression of Pcp4l1, Fcer1g, B4galt3, Ppox, Ufc1, Nit1, Usf1, Copa, Pex19, Wdr42a, Igsf8, Dfy, Fmn2 and Rgs7.
Analysis of six BXD recombinant inbred strains further refined Qrr1 into proximal and distal segments named Qrr1p and Qrr1d, respectively. Qrr1p spans 172 Mb (92.3 cM) to 174.5 Mb (95 cM) while Qrr1d spans 174.5 Mb (95 cM) to 177.5 Mb (99.8 cM). High-ranking candidate genes for Qrr1p include Pcp4l1, Pfdn2, Wdr42a and Kcnj10 while high-ranking for Qrr1d include Fmn2 and Rgs7.
