Linkage analysis was performed using a backcross derived from closely related inbred strains DBA/1J and DBA/2J. The genetic composition of these 2 inbred strains differs by just 5.6% at the SNP level. A total of 294 mice from (DBA/1J x DBA/2J)F1 x DBA/1Jand (DBA/1J x DBA/2J)F1 x DBA/2J populations were screened for 6,949 polymorphic SNP markers. Backcross mice were placed on a high-fat diet at 8 weeks of age and phenotyped for plasma triglyceride and HDL concentrations at 8, 11 and 16 weeks of age. Parental strain DBA/2J displays significantly increased plasma triglyceride concentrations on a CHOW diet compared to parental strain DBA/1J. Male DBA/2J animals display significantly increased plasma triglyceride and HDL concentrations after an 8 week high fat diet compared to DBA/1J males.

Tgq18 (triglyceride QTL 18) mapped to 40 cM on mouse Chromosome 18 at rs13479852 (82.89 Mb) with LOD=4.5. DBA/2J-derived alleles at Tgq18 confer increased plasma triglyceride concentrations on a CHOW diet. This locus also displays suggestive linkage to plasma triglyceride levels after 3 weeks on a high fat diet (LOD=2.7) and after 8 weeks on a high fat diet (LOD=2.9). [Table 2 and 3].
