Congenic animals carrying C57BL/6J-derived DNA at the Alcw2 locus (40 cM on chromosome 4) on a DBA/2J genetic background were used to study the effects of ethanol withdrawal on neural circuitry. The congenic line is designated D2.B6-Alcw2C57BL/6J. Parental strain DBA/2J exhibits greater alcohol withdrawal severity compared to C57BL/6J.

Expression of c-Fos was used as a marker of neuronal activation and was consistently increased in 24 out of 25 brain regions of the extended basal ganglia during ethanol withdrawal. However, D2.B6-Alcw2C57Bl/6J congenic animals display decreased ethanol withdrawal severity concomitant with significantly decreased c-Fos positive neurons in six brain regions during ethanol withdrawal compared to animals from the DBA/2Jbackground strain. These regions are the ventral pallidum (VP), rostromedial lateral globus pallidum (LGP), medial globus pallidus (MGP), subthalamic nucleus (STN) and substantia nigra pars reticulata (SNr). Mpdz (38.6 cM) is a potential candidate gene for Alcw2.
