    A previously identified QTL for haloperidol-induced catalepsy, Hpic2 (haloperidol induced catalepsy 2), on distal mouse Chromosome 1 was confirmed using congenic line B6.D2-Mtv7a/TyJ, which carries DBA/2J-derived DNA from D1Mit145 (89 cM; 169.1 Mb) to D1Mit17 (106.3 cM; 191.3 Mb) on a C57BL/6J genetic background. B6.D2-Mtv7a/TyJ and DBA/2J donor strain mice are sensitive to haloperidol-induced catalepsy. Fifteen minutes after haloperidol injection the animals were placed in fixed rearing position. Catalepsy-sensitive animals maintained the rearing position for over 30 seconds. C57BL/6J animals do not display this catalepsy response after haloperidol administration.

    The Hpic2 locus was refined further using a series of interval specific congenic lines derived from B6.D2-Mtv7a/TyJ backcrossed to C57BL/6J and selecting for recombination across the congenic interval. This method localized Hpic2 to a 5.9 Mb interval between D1Mit403 (100 cM; 177.5 Mb) to D1Mit273 (102.5 cM; 183.4 Mb) with the highest association to haloperidol sensitivity at 101 cM at D1Mit426 (P=0.0001). This interval contains 53 known and predicted genes.

    A screen for candidate genes with non-conservative amino acid changes and whole brain and/or striatum mRNA expression differences betweenC57BL/6J and DBA/2J was performed. Ephx1(98.5 cM) is a possible candidate gene for Hpic2 as it contains a non-conservative SNP, displays differential mRNA expression between C57BL/6J and DBA/2J, and is a cis-eQTL for haloperidol sensitivity. Lin9 (182.5 Mb) is a possible candidate with non-conservative SNP variations between C57BL/6J and DBA/2J. Cabc1 (182.09 Mb) is also a potential candidate for Hpic2, with differentially expressed striatum mRNA and weak linkage as a cis-eQTL. Potential candidate genes identified solely by mRNA expression variations between C57BL/6J and DBA/2J whole brain and striatal mRNA include Tmem63a (182.87 Mb), Parp1 (98.6 cM; 182.49 Mb), Psen2 (101 cM; 182.15 Mb), Nvl (183.02 Mb) and Cnih4 (183.08 Mb).
