Somatic cell hybrid line ACH was from fusion of Chinese hamster Tk-1 deficient cell line a3 with macrophages of C3H/HeJ mice. Of 15 hybrid clones selected in HAT medium only line ACH 6 retained mouse chromosomes. Nine secondary clones of ACH 6 were grownin HAT medium, medium with BrdU or nonselective medium. Five of the nine subclones that were capable of growth in both HAT and the nonselective medium (Tk-1+) were also positive for Chromosome 11 gene Glk. Four of the nine clones that were backselected in BrdU showed no expression of Glk. The concordant segregation of Tk-1 and Glk activity suggest assignment of Tk-1 to Chromosome 11.
