  • Experiment
DNA from the spleen of a 129/Sv male mouse was cleaved with rare cutting enzymes to produce common fragments that could be recognized by probes for Hoxd4 and Hoxd3. Probe A is a 2kb fragment of cosmid E lying 8kb from the 3' end of Hoxd4 and probe B is a1.8kb fragment of a lambda genomic clone lying 8-10kb upstream from Hoxd3. Restriction analysis illustrate that probes A and B hybridize to common fragments of 90kb from ClaI and 100 and 130kb from SfiI and to larger fragments generated by MluI and NruI.When the position of these two probes were analyzed it was suggested that Hoxd4 and Hoxd3 are no greater than 60kb apart. A jumping clone, Je - Js defining HoxD4 and HoxD3, was employed to establish an intergene distance of 35kb between Hoxd4 and Hoxd3 on Chromosome 2.
