Notes
  • Experiment
Somatic cell hybrids that were propagated in HAT media were derived from fusions of heterozygous T(11;19)42H mouse fibroblast cells with rat cell line RT2 Tk-. The hybrids were constructed for the purpose of determining the locus for Tk1 relative to the T(11;19)42H breakpoint in band 11D. Southern analysis was performed on 10 hybrids using probes for Il3, Hba, Erba and P4hb all of which span the 11D breakpoint. Of the 10 hybrids, three showed hybridization to all four probes, seven showed hybridization to markers below 11D (Erba, P4hb) and no hybridization to markers above 11D (Il3, Hba). Because the Tk1 locus is selected for in HAT media, a regional assignment of Tk1 could be identified as distal to the T42H breakpoint in band 11D.
