Experiment
Male and female C57BL/6J, A/J and AXB/BXA RI mice were used to map drug induced psychomotor actiavation using four breeder pairs/strain. All doses of alcohol, cocaine and morphine were administered intraperioneally
in volumes ranging from 0.005 to 0.02ml/g body weight. Activity was measured in open-field boxes. QTL analysis for morphine induced activity was measured in 342 mice (157 males and 185 females) from the AXB/BXA. Significant QTL were determined using maximum likelihood ratio statistics (LRS). Six loci were mapped for morphine induced locomoter activation, in parenthesis is the peak marker, cM location, and LRS score respectively. Chromosome 5, Dipa1 (D5Mit24, 60 cM, LRS 14.6), Chromosome 7, Dipa2 (D7Mit70, 27.8 cM, LRS 38.3) Chromosome11, Dipa 3(Pmv46, 11.0 cM, LRS 37.06), Chromosome 12, Dipa4 (D12Mit233, 52.0 cM, LRS 28.8), Chromosome 15, Dipa5 (D15Mit39, 56.6 cM, LRS 53.01) and Chromosome 17 Dipa6 (D17Mit18, 4.0 cM, LRS 38.21). Suggestive QTLs were found in Chromosome 1 (Iapls3-1 linked), Chromosome 3 (Mpmv9 linked), Chromosome 6(D6Mit368 linked) and Chromosome 12 (Pmv3 linked).
Joint morphine - cocaine - alcohol activation was also analyzed where in two chromosome regions were significant. Dipa7(Pmv46, 11.0 cM, LRS 34.10), Dipa8(D12Mit110, 19.0 cM, LRS 20.59). Suggestive QTLs were found in Chromosome 5 (D5Mit24 linked), Chromosome 8 (D8Mit224 linked), Chromosome 13(D13Mit115 linked) and Chromosome 15 (D15Mit28 linked).
