The authors describe the derivation of a diabetes resistant mouse strain, NOR/Lt, used in the identification of a new insulin-dependent diabetes susceptibility locus (Idd13) on Chromosome 2. NOR/Lt is a recombinant congenic strain in which portions of the NOD/Lt genome has been replaced by DNA from the C57BL/KsJ strain. Genotypic analysis of the progeny from a (NOD/Lt x NOR/Lt)F2 cross using the Chi square test indicates that Idd13 maps between B2m and Pcna, very near Il1. This location was confirmed by the analysis of congenic strains where Idd13 from the NOR/Lt strain cosegregates with Il1, D2Mit47 and D2Mit48 onto the NOD/Lt background. Together these studies indicate that the order of loci in this region of Chromosome 2 is: B2m, Idd13, Il1, D2Mit47, D2Mit48. Homozygosity for NOR/Lt-derived alleles confer resistance to IDDM at Idd13.
