Mice carrying only partial t haplotypes were analyzed for Iph1 expression. Sperm from mice homozygous for th49, carrying inversions In(17)1t and In(17)2t were analyzed and shown to only have the unphosphorylated form of HK1. This suggested that Iph1 is located in the proximal (in relation to the centromere) half of the t haplotype. Analysis of sprem from tTuw23/th2, which are homozygous for In(17)2t and heterozygous for In(17)1t, showed that HK1 is phosphorylated on tyrosine residues. Additionally, sperm HK1 from th2/th2 mice, homozygous for In(17)2t, was phosphorylated on tyrosine residues. Since mice homozygous for In(17)2t but carrying only one or no copies of In(17)1t had the tyrosine phosphorylated form of HK1, the Iph1 locus is not located in In(17)2t, and is therefore most likely located in In(17)1t on mouse Chromosme 17.
