  • Experiment
    CXXB cell lines were generated by transforming BM (Bromelain-treated mouse erythrocytes) cells from 8-week old (BALB/cAnn.xid x C57BL/10J)F1 mice with Abelson murine leukemia virus. Combined deletion mapping profiles made by digesting CXXB line DNA with EcoRI (probed with CH34VHPCR3.3 for Igh-V11) and XbaI (probed with CH27VHPCR90-5 for Igh-V12) defined the Igh variable region. Results showed two Igh-V11 haplotype genes 3' of genes in the Igh-VGAM3.8 family and 5' of two Igh-VS107 genes. A single Igh-V12gene mapped 3' to some genes of the Igh-VJ505 family and 5' to genes of the entire IGH-VH3660 family. The overall order of genes was reported on mouse Chromosome 12 as: Igh-VJ558 - Igh-V3609 - Igh-V606 - Igh-V12 - Igh-V3660 - Igh-VGAM3.8 - Igh-V3660 - Igh-VS107 - Igh-VGAM3.8 - Igh-V11 - Igh-V3660 - Igh-V11 - Igh-VX24 - Igh-VS107 - Igh-VQ52 - Igh-V7183 - Igh-D - Igh-J - Igh-C where repeats of genes represent the location of various haplotypes (family genes).
