Lyst Lystbg visible phenotype
Notes
  • Experiment
A YAC complementation strategy was used to localize the Lystbg locus in mice. A series of overlapping YACs (isolated by PCR screening for Nid sequences, previously shown to be near Lystbg on Mouse Chromosome 13) were introduced to bg mouse fibroblasts by spheroplast fusion. Fusions were screened for phenotypically small lysosomes no longer clustered in the perinuclear region and the disappearance of tubular lysosomes, indicating bg complementation. Complementary YACs were isolated and partially sequenced. The YACs were physically mapped using STS PCR analysis. Locus Lystbg was localized to a 500kb region roughly 70kb from Nid. The following order of loci on mouse Chromosome 13 was reported: D13Uth1 - D13Uth2 - Nis - D13Uth3 - D13Uth4 - Lyst - D13Uth5.
