Authors suggest that Om may be a candidate gene for rmy.
  • Experiment
As a preliminary analysis, two separate DNA pools were made from all of the mutant and wild-type progeny of the C3H/He-rmy/rmy x (C3H/He-rmy/rmy x C57BL/6J)F1 backcross. Enrichment or diminution was investigated for C3H and C57BL alleles for each MIT marker tested in the mutant DNA pool in comparison with the wild-type DNA pool. The two parental strains and the F1 hybrid were used as controls. Eight microsatellite markers were tested on Chromosome 11 to 13. For a Chromosome 11 marker, D11Mit20, a diminution of the C57BL allele was clearly visible in the mutant DNA pool as compared with the wild-type DNA pool and the F1 hybrid. Another Chromosome 11 marker, D11Mit132, showed a strong enrichment of the C3H allele and no visible C57BL allele in the mutant DNA pool. Authors state that this evidence shows linkage of these MIT markers to rmy and that these results strongly suggest that rmy maps to mouse Chromosome 11.
