    The Diversity Outbred heterogeneous stock (J:DO) is a developing mouse population derived from progenitor lines of the Collaborative Cross (CC). The CC is a panel of recombinant inbred (RI) mouse strains that combines the genomes of eight genetically diverse founder strains - A/J, C57BL/6J, 129S1/SvImJ, NOD/ShiLtJ, NZO/HlLtJ, CAST/EiJ, PWK/PhJ, and WSB/EiJ - to capture nearly 90% of the known variation present in laboratory mice (Churchill et al. 2004). Animals from 160 incipient CC lines at early stages of inbreeding were used to establish the DO population, which is maintained by a randomized outbreeding strategy that avoids brother-sister matings. The DO and CC populations thus capture the same set of natural allelic variants derived from a common set of eight founder strains, with DO mice being outbred and the CC population being inbred. Each CC inbred strain represents a fixed and reproducible genotype; each DO animal is a unique, non-reproducible individual with an effectively limitless combination of segregating alleles.

  • Experiment
    Linkage analysis was performed on a mapping population of 283 male and female J:DO mice (G4 and G5) to identify 5 novel loci related to various behavioral traits. SNP genotyping was performed utilizing the Mouse Universal Genotyping Array (MUGA), an Illumina Infinium Platform array with 7,851 SNP markers.

    QTL Ofcts is significant for the trait "open field - center time slope." It maps to 147.68 - 149.29 Mb on Chromosome 4 with a peak LOD score of 8.2. The PWK/PhJ allele is associated with decreased time spent in the center of the open-field arena, which is consistent with less time spent in the center of the open field by the PWK/PhJ progenitor strain.

    QTL Ofdi1 is significant for the trait "open field - duration immobile." It maps to 93.2 - 100.21 Mb on Chromosome 2 with a peak LOD score of 7.82. The allele effect plots indicate that NZO/H1LtJ alleles on chromosome 2 are associated with increased immobility in the open-field.

    QTL Ofdi2 is significant for the trait "open field - duration immobile." It maps to 114.07 - 115.94 Mb on Chromosome 6 with a peak LOD score of 8.63. CAST/EiJ alleles on chromosome 6 are associated with decreased immobility in the open-field.

    QTL Anxldb31 is significant for the trait "light-dark box - duration in light side (slope)." It maps to 95.01 - 96.55 Mb on Chromosome 11 with a peak LOD score of 7.4. The 129S1/SvlmJ allele is associated with a pronounced decrease in time spent in the light.

    QTL Tstc is significant for the trait "tail-suspension test - climbing behavior." It maps to 97.77 - 99.17 Mb on Chromosome 6 with a peak LOD score of 8.69. PWK/PhJ alleles are associated with an increase in climbing frequency.
