Linkage analysis was performed on two mapping populations of high-frequency (HF) and all-frequency (AF) hearing loss (HL) mice to identify loci related to hearing loss in mice:

    1) (AFHL x C3HeB/FeJ)F1 x AFHL (referred to as AFHL-N2 mice)

    2) (HFHL x C3HeB/FeJ)F1 x HFHL (referred to as HFHL-N2 mice)

Both the F1 crosses and the backcrosses were done reciprocally. 300 AFHL-N2 mice were genotyped at 257 SNP markers. 306 HFHL-N2 mice were genotyped at 145 SNP markers. Seven hearing loss QTL reaching genome-wide significance were identified:

QTL Hfhl1 (high-frequency hearing loss 1) was identified in AFHL-N2 mice and is significant for hearing loss measured by response to a 32 kHz tone pip. It maps to Chromosome 7 between the SNPs rs31944466 and rs3700384 with a peak LOD score of 3.3 at 54.89 Mb (rs31944466).

QTL Hfhl2 (high-frequency hearing loss 2) was identified in AFHL-N2 mice and is significant for hearing loss measured by response to a 32 kHz tone pip. It maps to Chromosome 8 with a peak LOD score of 4.6 at 95.75 Mb (rs13479916).

QTL Hfhl3 (high-frequency hearing loss 3) was identified in HFHL-N2 mice and is significant for hearing loss measured by response to a 32 kHz tone pip. It maps to Chromosome 7 with a peak LOD score of 5.9 at 63.73 (rs6160140).

QTL Hfhl4 (high-frequency hearing loss 4) was identified in AFHL-N2 mice and is significant for hearing loss measured by response to a 32 kHz tone pip. It maps to Chromosome 10 with a peak LOD score of 40.0 at 70.85 Mb (rs6326263).

QTL Clfrhl1 (click-frequency hearing loss 1) was identified in AFHL-N2 mice and is significant for hearing loss measured by response to a click stimulus. It maps to Chromosome 10 with a peak LOD score of 93.8 at 75.67 Mb (rs13480652).

QTL Lfhl1 (low-frequency hearing loss 1) was identified in AFHL-N2 mice and is significant for hearing loss measured by response to an 8 kHz tone pip. It maps to Chromosome 10 with a peak LOD score of 91.3 at 75.67 Mb (rs13480652).

QTL Mfhl1 (medium-frequency hearing loss 1) was identified in AFHL-N2 mice and is significant for hearing loss measured by response to a 16 kHz tone pip. It maps to Chromosome 10 with a peak LOD score of 98.8 at 75.67 Mb (rs13480652).
