A startle reflex in response to an intense acoustic stimulus is inhibited when a barely detectable pulse precedes the startle stimulus by 30500 ms. It has been theorized that this phenomenon, named prepulse inhibition (PPI) of a startle response, is an automatic early-stage gating process contributing to the ability to focus attention. Deficits in PPI may therefore contribute to deficits in attentional processing. Both deficits are observed in schizophrenia spectrum disorders. The authors investigated whether there is overlap in genetic control of PPI and attentional processing phenotypes in the panel of BXD recombinant inbred (RI) strains of mice.

Linkage analysis was performed on a mapping population of 422 mice from 35 BXD RI strains to identify QTL related to PPI. Mapping was performed using GeneNetwork's embedded MapManager software (BXD RI SNP data included). The search identified three QTL reaching genome-wide significance (NCBI Build 37):

QTL Ppipq1 (prepulse inhibition percentage QTL 1) maps to Chromosome 17 from 71.1 - 75.47 Mb (peak marker and LOD score not given). The peak additive effect in terms of PPI was 7.63%, with the C57BL/6J allele contributing to higher PPI scores at the Ppipq1 locus than the DBA/2J allele.

QTL Vasr1 (variability in acoustic startle response 1) maps to Chromosome 16 from 19.88 - 26.55 Mb (peak marker and LOD score not given). The C57BL/6J allele contributes to reduced response variability (i.e. increased attentional performance) at the Vasr1 locus.

QTL Masrl1 (mode of acoustic startle response latencies 1) maps to Chromosome 16 from 93.93 - 97.54 Mb (peak marker and LOD score not given). No allelic information was given for this locus.
