Enzyme activity is 2.2-fold greater in brain extracts of targeted mutant/transgenic (Tg888) than of wild-type mice. (As Tg890 has only a 6% elevation over wild-type brain enzyme activity, Tg888 was used for all further analyses.) Immunoperoxidase staining of brain sections showed expression in astrocytes, but not neurons, of wild-type brain and essentially uniform expression throughout the brain. In contrast, targeted mutant/transgenic brains exhibited stronger but diffuse expression in both neurons and astrocytes and greater, consistent variation of expression among different regions, with especially high expression in pyramidal neurons of the hippocampus. Indirect electrophysiologic assays of stimulation via the adenosine A1 receptor in brain sections demonstrated diminished ambient adenosine levels in CA3 hippocampal pyramidal neurons of transgenic mice.
Mice homozygous for the Adktm1Bois null mutation and heterozygous for the transgene exhibit rescue of the lethality and the steatotic liver phenotype associated with adenosine kinase deficiency. |
