Temt) was proposed as a novel candidate gene for previously identified QTL Srilp (sleep response to influenze, light phase) on mouse Chromosome 6. Gene expression analysis of influenza-infected animals showed significantly varied dark-light phase Inmt mRNA expression in the hypothalamus and basal forebrain of high sleep response strain C57BL/6J, but not in low sleep response strain BALB/cByJ. Expression of several other Srilp candidate genes (Ghrhr, Crhr2, Npy, Adcyap1r1) were also examined but none were expressed differently between influenza-infected BALB/cByJ and C57BL/6J animals. Sequence analysis of Inmt revealed a conservative single nucleotide substitution at position 95. However, the strain distribution pattern of this polymorphism does not correlate with the Srilp phenotype. Several other intronic SNPs were also identified in Inmt and Crhr2 sequences. Authors suggest it is unlikely that Inmt is the gene underlying Srilp. However, the 95% confidence interval of Srilp was localized to 28 cM - 31 cM. Srilp accounts for 35%-45% of the phenotypic variance. |
