Mutation detailsA vector pBI-G (Clontech #6150-1) was used to create this transgene. The vector contains a prokaryotic beta-galactosidase (lacZ) reporter gene under the control of the tetracycline-response element (TRE; containing seven direct repeats of the tetracycline operator (tetO) sequence fused to bidirectional cytomegalovirus (CMV) minimal promoter). The rat Aurora-kinase B cDNA (with N-terminal FLAG-tag sequence and 3' linked beta-globin gene) was placed downstream of the promoter and the purified construct was injected into fertilized FVB/NJ eggs to create the transgenic mice.
