Mutation details: This transgene contains the tet operon (tetO) and a minimal CMV promoter controlling expression of a cDNA encoding a dominant-negative mutant rat cAMP-responsive element binding protein 1 (CREB1), alpha isoform, bearing a FLAG peptide tag (Asp-Tyr-Lys-Asp-Asp-Asp-Asp-Lys). The coding sequence is followed by the SV40 intron and polyadenylation signal. The mutant CREB1 has alanine in place of the critical regulatory serine at position 133, eliminating the phosphorylation site. Unphosphorylated CREB binds to cAMP response elements (CREs), blocking access by wild-type CRE and other CRE-binding proteins.
