Mutation detailsA BAC was modified to truncate murine Pkd1 such that the region just prior to the first transmembrane and the membrane-spanning and intracellular domains is deleted and produces only the extracellular domain. A second modification removed the Tsc2 gene from the BAC. Immunoblotting confirmed protein expression in various tissues including kidney, brain, liver, lung, and spleen. Four lines were generated (2, 9, 11, and 39) with 2 to 85 copies of the transgene. All lines show a similar phenotype. Line 39 is the low expressing line with 2 copies of the transgene. (J:198147)
