Mutation detailsThe 130 kb YAC transgene, YZ, contains the Igf2 and H19 genomic regions and in which the Igf2 coding sequence was interrupted with the lacZ reporter gene. The YZ YAC extends from promoter 1 of Igf2 to 35 kb downstream of H19, and includes the differentially methylated region around exons 5 to 6 of Igf2, the unmethylated intergenic hypersensitive region, A6-A4, the differentially methylated regions around H19 and the endoderm-specific enhancers downstream of H19. LacZ activity shows spatial expression of Igf2 in a subset of tissues. The imprinted H19 gene is only expressed when transmitted through the maternal germline. After paternal transmission the gene is repressed. Line YZ8 carries a single copy of the transgene. (J:43322)
